Clerks 2: No. Look for an animated feature at some point, but no Clerks 2.

The Relationship: They all take place in the same universe, where certain parts and characters are repeated/have an effect on events in the other movies. Want a better answer? Read the summaries.

Clerks, Mallrats, and the time span: Read the summaries. I've always assumed that the events in Mallrats occur after Clerks, however, but I'm sure you can find evidence either way.
