That said, here's a codicil:
Depends on the goal of the comedic material. Do what you find funny, find an audience that appreciates your take on things, and you're home free.
Sometimes, though, you have to consider the makeup of the audience/reader.
Some comedy, while inherantly funny in and of itself, will not be so to certain groups.
At times, that can be half the fun: ex) I do a Witch Trial show which is designed to be unfunny for bookburning fundamentalists (as they and their ilk are being mocked throughout). It's a hoot and a half for the thinkers in the crowd, but some folks just stare at me like a dog being shown a card trick...but that's only to be expected, as fundamentalism (of whatever theism) tends to breed a lack of irony and the inability to perceive same.
